RESUMO

Evidence suggests that tart cherry (TC) supplementation has beneficial effects on health indices and recovery following strenuous exercise. However, little is known about the mechanisms and how TC might modulate the human metabolome. The aim of this study was to evaluate the influence of an acute high- and low-dose of Vistula TC supplementation on the metabolomic profile in humans. In a randomised, double-blind, placebo controlled, cross-over design, 12 healthy participants (nine male and three female; mean ± SD age, stature, and mass were 29 ± 7 years old, 1.75 ± 0.1 m, and 77.3 ± 10.5 kg, respectively) visited the laboratory on three separate occasions (high dose; HI, low dose; LO, or placebo), separated by at least seven days. After an overnight fast, a baseline venous blood sample was taken, followed by consumption of a standardised breakfast and dose conditions (HI, LO, or placebo). Subsequent blood draws were taken 1, 2, 3, 5, and 8 h post consumption. Following sample preparation, an untargeted metabolomics approach was adopted, and the extracts analysed by LCMS/MS. When all time points were collated, a principal component analysis showed a significant difference between the conditions (p < 0.05), such that the placebo trial had homogeneity, and HI showed greater heterogeneity. In a sub-group analysis, cyanidine-3-O-glucoside (C3G), cyanidine-3-O-rutinoside (C3R), and vanillic acid (VA) were detected in plasma and showed significant differences (p < 0.05) following acute consumption of Vistula TC, compared to the placebo group. These results provide evidence that phenolics are bioavailable in plasma and induce shifts in the metabolome following acute Vistula TC consumption. These data could be used to inform future intervention studies where changes in physiological outcomes could be influenced by metabolomic shifts following acute supplementation.

Smokeless powders (SLPs) are composed of a combination of thermolabile and non-thermolabile compounds. When analysed by GC-MS, injection conditions may therefore play a fundamental role on the characterisation of forensic samples. However, no systematic investigations have ever been carried out. This casts doubt on the optimal conditions that should be adopted in advanced profiling applications (e.g. class attribution and source association), especially when a traditional split/splitless (S/SL) injector is used. Herein, a study is reported that specifically focused on the evaluation of the liner type (Ltype) and inlet temperature (Tinj). Results showed that both could affect the exhaustiveness and repeatability of the observed chemical profiles, with Ltype being particularly sensitive despite typically not being clarified in published works. Perhaps as expected, degradation effects were observed for the most thermolabile compounds (e.g. nitroglycerin) at conditions maximising the heat transfer rates (Ltype = packed and Tinj ≥ 200 °C). However, these did not seem to be as influential as, perhaps, suggested in previous studies. Indeed, the harshest injection conditions in terms of heat transfer rate (Ltype = packed and Tinj = 260 °C) were found to lead to better performances (including better overall %RSDs and LODs) compared to the mildest ones. This suggested that implementing conditions minimising heat-induced breakdowns during injection was not necessarily a good strategy for comparison purposes. The reported findings represent a concrete step forward in the field, providing a robust body of data for the development of the next generation of SLP profiling methods.

We report, for the first time, a multimodal investigation of current crude oil reprocessing and storage sites to assess their impact on the environment after 50 years of continuous operation. We have adopted a dual approach to investigate potential soil contamination. The first approach uses conventional analytical techniques i.e. energy dispersive X-ray fluorescence (ED-XRF) for metal analysis, and a complementary metabolomic investigation using hydrophilic liquid interaction chromatography hi-resolution mass spectrometry (HILIC-MS) for organic contaminants. Secondly, the deployment of an unmanned aerial vehicle (UAV) with a multispectral image (MSI) camera, for the remote sensing of vegetation stress, as a proxy for sub-surface soil contamination. The results identified high concentrations of barium (mean 21 017 ± 5950 µg g-1, n = 36) as well as metabolites derived from crude oil (polycyclic aromatic hydrocarbons), cleaning processes (surfactants) and other organic pollutants (e.g. pesticides, plasticizers and pharmaceuticals) in the reprocessing site. This data has then been correlated, with post-flight data analysis derived vegetation indices (NDVI, GNDVI, SAVI and Cl green VI), to assess the potential to identify soil contamination because of vegetation stress. It was found that strong correlations exist (an average R2 of >0.68) between the level of soil contamination and the ground cover vegetation. The potential to deploy aerial remote sensing techniques to provide an initial survey, to inform decision-making, on suspected contaminated land sites can have global implications.

The flowering stage of oilseed rape (Brassica napus L.) is of vital interest in precision agriculture. It has been shown that data describing the flower production of oilseed rape (OSR), at stage 3, in spring can be used to predict seed yield at harvest. Traditional field-based techniques for assessing OSR flowers are based on a visual assessment which is subjective and time consuming. However, a high throughput phenotyping technique, using an unmanned aerial vehicle (UAV) with multispectral image (MSI) camera, was used to investigate the growth stages of OSR (in terms of crop height) and to quantify its flower production. A simplified approach using a normalised difference yellowness index (NDYI) was coupled with an iso-cluster classification method to quantify the number of OSR flower pixels and incorporate the data into an OSR seed yield estimation. The estimated OSR seed yield showed strong correlation with the actual OSR seed yield (R2 = 0.86), as determined using in-situ sensors mounted on the combine harvester. Also, using our approach allowed the variation in crop height to be assessed across all growing stages; the maximum crop height of 1.35 m OSR was observed at the flowering stage. This methodology is proposed for effectively predicting seed yield 3 months prior to harvesting.

Laundering of textiles is a significant source of waterborne microfiber pollution, and solutions are now being sought to mitigate this issue including improvements in clothing technology and integration of filtration systems into washing machines. Vented tumble dryers are a potential source of airborne microfiber pollution, as their built-in lint filtration systems have been found to be inefficient with significant quantities of textile microfibers being released to the external environment through their exhaust air ducts. The present study is the first to evaluate the impact of condenser dryers, finding that they are significant contributors to waterborne microfiber pollution from the lint filter (if users clean this with water), the condenser and the condensed water. Microfiber release from drying of real consumer loads in condenser and vented tumble dryers was compared, finding that real loads release surprisingly high levels of microfibers (total 341.5 ± 126.0 ppm for those dried in a condenser dryer and 256.0 ± 74.2 ppm for those dried in a vented dryer), similar in quantity to microfibers produced during the first highly-shedding drying cycle of a new T-shirt load (total 321.4 ± 11.2 ppm) in a condenser dryer. Vented dryers were found to be significant contributors to waterborne microfiber pollution if consumers clean the lint filter with water in accordance with some published appliance usage instructions, as most (86.1 ± 5.5% for the real consumer loads tested) of the microfibers generated during vented tumble drying were collected on the lint filter. Therefore, tumble dryers are a significant source of waterborne and (for vented dryers) airborne microfiber pollution. While reducing the pore size of tumble dryer lint filters and instructing consumers to dispose of fibers collected on lint filters as municipal solid waste could help reduce the issue, more sophisticated engineering solutions will likely be required to achieve a more comprehensive solution.

Vented tumble dryers release moist warm air from the drying process to the external environment, usually through pipework linking the appliance to a vent in an exterior wall. Although such dryers contain a lint filter to remove fibers from this air stream, recent reports suggest that this process is incomplete, leading to microfibers being released in the ducted warm air and subsequently polluting the external environment. Microfiber release from wash loads comprising 10 100% cotton and 10 100% polyester T-shirts (total load mass ratio 48% cotton, 52% polyester) was measured at different stages of the washing and drying process to compare the quantities of fibers released 'down the drain', collected in the dryer lint filter, and released to air from the tumble dryer. Testing under both European and North American washing conditions found that the quantities of microfibers released to air during tumble drying were significant and comparable to levels released 'down the drain' during washing. Use of conventional rinse-added liquid fabric conditioner increased microfiber accumulation on the dryer lint filter, with reduced release from the dryer exhaust observed at the highest fabric conditioner dose tested (21.6% and 14.2% reduction under North American and European conditions, respectively). Conventional liquid fabric conditioner did not significantly impact microfiber release from the washing machine, in line with previous studies. A fabric conditioner specially designed for anti-wrinkle performance reduced microfiber release from the dryer exhaust at all levels tested (by 17.6-35.6%, depending on dose), apparently by increasing the efficiency of microfiber accumulation in the lint filter. Tumble dryer sheets were also found to cause a reduction in microfiber release from the dryer exhaust (by 14.1-34.9%, depending on the dose/product), likely driven by collection of liberated fibers on the sheet during the drying process. The use of both antiwrinkle liquid fabric conditioner and dryer sheet enabled a 44.9% reduction in microfiber emissions from the dryer exhaust. In all studies, the fiber mass collected on the lint filter or emitted from the dryer exhaust was richer in cotton fibers (range 83.4-96.3% on the lint filter, 93.0-99.8% from the dryer exhaust) than the wash load composition (48% cotton). Moreover, fibers collected by the lint filter contained a higher proportion of polyester than emissions from the dryer exhaust (range 3.7-16.6% on the lint filter, 0.2-7.0% from the dryer exhaust). There is significant variation in the porosity of lint filters among installed vented tumble dryers. Single-variable testing of the impact of lint filter design concluded that reducing screen pore size significantly reduces airborne microfiber release during tumble drying; a reduction in lint filter pore size from 0.2 mm2 to 0.04 mm2 reduced release by 34.8%. As some lint filters have pore sizes of around 1 mm2, there is enormous scope to reduce microfiber release from dryers though improved lint filter design. However, it is suggested that a step-change in appliance design away from vented dryers to only fully-sealed condenser dryers might be necessary to eliminate the contribution of tumble drying to airborne microfiber pollution.

An area of ancient and semi-natural woodland (ASNW) has been investigated by applied aerial spectroscopy using an unmanned aerial vehicle (UAV) with multispectral image (MSI) camera. A novel normalised difference spectral index (NDSI) algorithm was developed using principal component analysis (PCA). This novel NDSI was then combined with a simple segmentation method of thresholding and applied for the identification of native tree species as well as the overall health of the woodland. Using this new approach allowed the identification of trees at canopy level, across 7.4 hectares (73,934 m2) of ASNW, as oak (53%), silver birch (37%), empty space (9%) and dead trees (1%). This UAV derived data was corroborated, for its accuracy, by a statistically valid ground-level field study that identified oak (47%), silver birch (46%) and dead trees (7.4%). This simple innovative approach, using a low-cost multirotor UAV with MSI camera, is both rapid to deploy, was flown around 100 m above ground level, provides useable high resolution (5.3 cm / pixel) data within 22 mins that can be interrogated using readily available PC-based software to identify tree species. In addition, it provides an overall oversight of woodland health and has the potential to inform a future woodland regeneration strategy.

The opium alkaloids (morphine, codeine, thebaine, noscapine, and papaverine) have been detected on poppy seeds; they are widely used by the food industry for decoration and flavor but can introduce opium alkaloids into the food chain. Of the opium alkaloids found on poppy seeds, morphine, and codeine are the most pharmacologically active and have been detected in biological matrices collected in workplace and roadside drug testing resulting in positive opiate results. The European Food Safety Authority introduced an acute reference dose of 10 µg morphine/kg of body weight as a safe level for morphine in food products. In this work, it was found that in harvested poppy seeds, and thermally processed poppy seeds (with and without a food matrix), if used in normal levels would not exceed the recommended acute reference dose. It was also shown that the levels of all alkaloids reduce when thermally processed, in comparison with harvested, untreated seeds.

Global concern exists regarding human exposure to organic pollutants derived from public open spaces and indoor dust. This study has evaluated the occurrence of 18 polycyclic aromatic hydrocarbons (PAHs), 11 organophosphorus flame retardants (OPFRs) and bisphenol A (BPA). To achieve this, a new simple, efficient and fast multi-residue analytical method based on a fully automated pressurised liquid extraction (PLE) and subsequent quantification by gas chromatography coupled to electron ionization-mass spectrometry (GC-EI-MS) in selected ion monitoring (SIM) mode was developed. The developed method was applied to indoor dust (12 sampling households) and soil derived from two public open spaces (POSs). Among all compounds studied, PAHs were the most ubiquitous contaminants detected in POS soils and indoor dust although some OPFRs and BPA were detected in lower concentrations. An assessment of the incremental lifetime cancer risk (ILCR) was done and indicated a high potential cancer risk from the POS sites and some of the indoor dust sampled sites. However, key variables, such as the actual exposure duration, frequency of contact and indoor cleaning protocols will significantly reduce the potential risk. Finally, the ingestion of soils and indoor dust contaminated with OPFRs and BPA was investigated and noted in almost all cases to be below the USEPA reference doses.

With the now widescale reporting of oral bioaccessibility data at contaminated sites, following our investigation of three sites (one public open space and two residential) for As and Pb contamination, a critical evaluation of the application and utility of such bioaccessibility testing was undertaken to better inform future use. Mean As and Pb soil levels across the sites varied between 12.5 and 24,900 mg/kg and 149-5930 mg/kg, respectively. Using the Unified Bioaccessibility Method (UBM) for in vitro bioaccessibility testing the highest bioaccessible concentrations were identified in the gastric phase. At site 1, a residential urban garden site the maximum bioaccessible As was 50.2% while the maximum bioaccessible Pb was 64.8%; similarly in site 2, also a residential urban garden site the maximum bioaccessible As was 38.72% while the maximum bioaccessible Pb was 66.0%. However, at site 3, a public open space site, the maximum bioaccessible As was 29.7% while the maximum bioaccessible Pb was 38.4%. Using the appropriate soil screening values and recommended statistical testing, we highlight that the use of bioaccessibility testing was unnecessary at sites 1 and 2 (residential urban garden sites), while at site 3 the value of oral bioaccessibility testing is highlighted as part of a 'lines of evidence approach' to support the site's specific risk assessment. We need to move away from the uncritical, blanket application of oral bioacessibility testing and strategically target where the results of these data add real value to site determination.

Fiber release during domestic textile washing is a cause of marine microplastic pollution, but better understanding of the magnitude of the issue and role of fabric care products, appliances and washing cycles is needed. Soiled consumer wash loads from U.K. households were found to release a mean of 114 ± 66.8 ppm (mg microfiber per kg fabric) (n = 79) fibers during typical washing conditions and these were mainly composed of natural fibers. Microfiber release decreased with increasing wash load size and hence decreasing water to fabric ratio, with mean microfiber release from wash loads in the mass range 1.0-3.5 kg (n = 57) found to be 132.4 ± 68.6 ppm, significantly (p = 3.3 x 10-8) higher than the 66.3 ± 27.0 ppm of those in the 3.5-6.0 kg range (n = 22). In further tests with similar soiled consumer wash loads, moving to colder and quicker washing cycles (i.e. 15°C for 30 mins, as opposed to 40°C for 85 mins) significantly reduced microfiber generation by 30% (p = 0.036) and reduced whiteness loss by 42% (p = 0.000) through reduced dye transfer and soil re-deposition, compared to conventional 40°C cycles. In multicycle technical testing, detergent pods were selected for investigation and found to have no impact on microfiber release compared to washing in water alone. Fabric softeners were also found to have no direct impact on microfiber release in testing under both European and North American washing conditions. Extended testing of polyester fleece garments up to a 48-wash cycle history under European conditions found that microfiber release significantly reduced to a consistent low level of 28.7 ± 10.9 ppm from eight through 64 washes. Emerging North American High-Efficiency top-loading washing machines generated significantly lower microfiber release than traditional top-loading machines, likely due to their lower water fill volumes and hence lower water to fabric ratio, with a 69.7% reduction observed for polyester fleece (n = 32, p = 7.9 x 10-6) and 37.4% reduction for polyester T-shirt (n = 32, p = 0.0032). These results conclude that consumers can directly reduce the levels of microfibers generated per wash during domestic textile washing by using colder and quicker wash cycles, washing complete (but not overfilled) loads, and (in North America) converting to High-Efficiency washing machines. Moving to colder and quicker cycles will also indirectly reduce microfiber release by extending the lifetime of clothing, leading to fewer new garments being purchased and hence lower incidence of the high microfiber release occurring during the first few washes of a new item.

A novel, rapid and sensitive analytical method has been developed and applied to 105 sputum samples from patients with cystic fibrosis, including 5 samples from post-lung transplant patients. This new method is specifically targeted to measure ß-alanyl aminopeptidase activity which is characteristic of some important Gram-negative pathogens. Of relevance to this study are Pseudomonas aeruginosa and pathogens of the Burkholderia cepacia complex both of which are commonly associated with respiratory infections as well as increased morbidity and mortality in adult cystic fibrosis patients. The analytical method involves the addition of a novel enzyme substrate (i.e. 3-amino-N-(3-fluorophenyl)propanamide) that interacts with ß-alanyl aminopeptidase to generate an exogenous volatile organic compound 3-fluoroaniline (LOD 0.02 µg mL-1; LOQ 0.06 µg mL-1). 3-Fluoroaniline was determined at 20 times above its calculated limit of quantification in the sputum samples by HS-SPME-GC-MS and then the results compared with standard culture methods and bacterial identification using MALDI-TOF-MS. Detection of 3-fluoroaniline was possible after only 8 h incubation of the sputum samples with a 95% success rate; this increased to 100% at 24 h which was well within the typical routine timeframe of 48 h. To our knowledge, this is the first demonstration of detection of P. aeruginosa by use of a custom-designed substrate to liberate a detectable and unique VOC. The very high negative predictive value (100% in this study) means such an assay could be appropriate as a screening technique for patients who are not yet colonized by this pathogen.

A new and improved method was developed for the determination and quantification of four "free" thyroid hormones (i.e. 3,5-diiodothyronine (T2), 3,3',5-triiodothyronine (T3), 3,3',5'-triiodothyrone (rT3) and 3,5,3',5'-tetraiodothyronine (T4)) in human serum by low- and high-resolution liquid chromatography-mass spectrometry (LC-MS). Several sample preparation strategies were investigated to obtain matrix-independent results. These strategies included solid phase extraction and matrix dilution. The developed analytical methods were then directly compared, in a blind study using patient-derived human blood serum samples, to the current clinical routine testing methods, i.e. electrochemiluminescence immunoassay and enzyme-linked immunosorbent assay. Chromatographic separation was achieved on a pentafluorophenyl (F5) column with an isocratic method of 30% aqueous phase, 70% organic phase where mobile phase A is 0.1% formic acid in water (pH 4) and mobile phase B is 0.1% formic acid in methanol (pH 4) (v/v). The high-resolution LC-MS was able to give a significant improvement in sensitivity with limits of quantification of 0.002 to 0.008 pmol/L for all four "free" thyroid hormones, as well as reduced sample preparation, making this the preferred method. However, the increase in capital cost may be beyond the capabilities of some laboratories. The LC-MS methods allow for the analysis of "free" thyroid hormones to be carried out in a significantly reduced analysis time. Clinical sample analysis showed that there was no statistical difference between the results obtained by ECLIA/ELISA and both LC-MS methods. Graphical abstract.

Globally, many of our urban agriculture sites (UAS) contain high levels of lead (Pb), a contaminant of toxicological concern to humans. To improve the derivation of soil assessment criteria at UAS, and avoid inappropriate closure of these valuable community spaces, we sampled nearly 280 paired soil and crop samples across 31 UAS gardens. This sampling was coupled with an exposure and food frequency questionnaire and participants blood Pb levels (BLL), (43 gardeners and 29 non-gardening neighbours). In 98% of the sampled soils, Pb concentrations were above the current UK soil guideline for UAS (80 mg/kg), however despite the high soil Pb (geometric mean: 324 mg/kg), and high soil bioaccessible Pb (geometric mean: 58.7%), all participants BLL were <4.1 µg/dL (range: 0.6-4.1 µg/dL). Indeed, there was no statistically significant difference between the BLL of the UAS gardeners and those of their non-gardening neighbours (p = 0.569). Pb uptake, however, varied with crop type and our study highlights the suitability of certain crops for growing at UAS with elevated Pb (e.g. tubers, shrub and tree fruit), whilst limiting the consumption of others (selected root vegetables, such as rhubarb, beetroot, parsnips and carrots, with observed Pb concentrations > 0.1 mg/kg FW). The importance of defining the exposure scenario of a specific sub-population (i.e. UAS gardeners) is highlighted. Our preferred models predict site specific assessment criteria (SSAC) of 722-1634 mg/kg. We found fruit and vegetable consumption rates by all participants, and not just the UAS gardeners, to be considerably higher than those currently used to derive the UK's category 4 screening levels (C4SLs). Furthermore, the soil to plant concentration factors (SPCFs) used to derive the UAS C4SL significantly over predict Pb uptake. Our study indicates it may be appropriate to develop a distinct exposure dataset for UAS. In particular we recommend the derivation of SPCFs that are reflective of urban soils, both in terms of the range of soil Pb concentrations typically observed, but also the sources (and hence human oral bioaccessibility and plant-availability) of this Pb.

Rapid, sensitive, and selective detection and identification of pathogenic bacteria is required in terms of food security. In this study, exogenous VOCs liberated by Salmonella strains have been identified and quantified via head space-solid phase microextraction gas chromatography mass spectrometry (HS-SPME-GC-MS) in milk samples. The specific enzymes targeted for detection and/or differentiation of Salmonella were C8 esterase, α-galactosidase and pyrrolidonyl peptidase using the following enzyme substrates: 2-chlorophenyl octanoate, phenyl α-d-galactopyranoside and L-pyrrollidonyl fluoroanilide, respectively. Detection of the exogenous VOCs, 2-chlorophenol, phenol and 3-fluoraniline was possible with typical limits of detection of 0.014, 0.045 and 0.005 µg/mL, respectively and correlation coefficients >0.99. The developed methodology was able to detect and identify Salmonella species within a 5 h incubation at 37 °C by the detection of the liberated VOCs. It was found that the milk samples tested were Salmonella free.

As part of a research programme to establish an analytical method for the simultaneous detection of the five major opium alkaloids in poppy seeds by liquid chromatography-electrospray ionization-mass spectrometry (LC-ESI-MS) it was discovered that the inclusion of thebaine produced two peaks for the same compound. This was in contrast to the effective simultaneous detection, by LC-ESI-MS, of morphine, codeine, papaverine and noscapine. The presence of these two peaks for thebaine was investigated using nuclear magnetic resonance spectroscopy with deuterated solvents to emulate the mobile phase conditions experienced. It was found that the presence of 80%, or higher ratios of, water caused two epimeric forms of thebaine to be formed; this explained the presence of two peaks on the chromatogram. In contrast, when a lower water content was used with 1% acetic acid, one stable form of thebaine could be analysed and resulted in a single peak visible in the subsequent chromatography.

The adverse health effects resulting from exposure to contaminated soil on internally displaced populations in Mitrovica, Kosovo can be determined by how the potentially harmful elements are bound in the soils. Certainly this was the case for Pb, present at concentrations ranging from 624 to 46,900 mg/kg, and at bioaccessibilities ranging <5% to nearly 90%. To assess why the soil Pb might differ so markedly in terms of its bioaccessibility, computer controlled scanning electron microscopy (CCSEM) was employed to determine how the Pb was associated with other elements at the individual particle (IP) level in soils from the area. It was found that the Pb-bearing particle types were, for the most part, different in each sample. We consider these differences as the main control on Pb bioaccessibility in these soils. Pb solubility at the IP level was evaluated by examining Pb-particles from these soils in the electron microscope before and after successive immersions in a simulated gastric fluid. This analysis (differential IP analysis) confirmed the CCSEM characterization that Pb associated with other higher atomic number elements (Fe, Zn, Cu and Ni) was less soluble than when it was present as isolated phases (e.g., as carbonate) or when it was bound with lower atomic number elements (Na, Al, Si, K, Ca). The heterogeneity in solubility and composition of the Pb-particles suggested that the Pb originated from a range of different anthropogenic activities. The nature of these different anthropogenic activities created the wide differences in Pb-bioaccessibilty by producing Pb bound in many different forms in the soil particles. This type of Pb-particle characterization highlights the role CCSEM analysis, and IP acid extraction, can play in providing supporting evidence alongside bioaccessibility data for applications in human health risk assessment and management of contaminated soil.

Listeria monocytogenes is a Gram-positive bacterium and an opportunistic food-borne pathogen which poses significant risk to the immune-compromised and pregnant due to the increased likelihood of acquiring infection and potential transmission of infection to the unborn child. Conventional methods of analysis suffer from either long turn-around times or lack the ability to discriminate between Listeria spp. reliably. This paper investigates an alternative method of detecting Listeria spp. using two novel enzyme substrates that liberate exogenous volatile organic compounds in the presence of α-mannosidase and D-alanyl aminopeptidase. The discriminating capabilities of this approach for identifying L. monocytogenes from other species of Listeria are investigated. The liberated volatile organic compounds (VOCs) are detected using an automated analytical technique based on static headspace-multi-capillary column-gas chromatography-ion mobility spectrometry (SHS-MCC-GC-IMS). The results obtained by SHS-MCC-GC-IMS are compared with those obtained by the more conventional analytical technique of headspace-solid phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS). The results found that it was possible to differentiate between L. monocytogenes and L. ivanovii, based on their VOC response from α-mannosidase activity.

This paper utilized L-alanine aminopeptidase activity as a useful approach to distinguish between Gram-negative and Gram-positive bacteria. This was done using two enzyme substrates, specifically 2-amino-N-phenylpropanamide and 2-amino-N-(4-methylphenyl)propanamide which liberated the volatile compounds aniline and p-toluidine, respectively. Two complementary analytical techniques have been used to identify and quantify the VOCs, specifically static headspace multicapillary column gas chromatography ion mobility spectrometry (SHS-MCC-GC-IMS) and headspace solid phase microextraction gas chromatography mass spectrometry (HS-SPME-GC-MS). Superior limits of detection were obtained using HS-SPME-GC-MS, typically by a factor of x6 such that the LOD for aniline was 0.02µg/mL and 0.01µg/mL for p-toluidine. In addition, it was also possible to determine indole interference-free by HS-SPME-GC-MS at an LOD of 0.01µg/mL. The approach was applied to a range of selected bacteria: 15 Gram-negative and 7 Gram-positive bacteria. Use of pattern recognition, in the form of Principal Component Analysis, confirmed that it is possible to differentiate between Gram-positive and Gram-negative bacteria using the enzyme generated VOCs, aniline and p-toluidine. The exception was Stenotrophomonas maltophilia which showed negligible VOC concentrations for both aniline and p-toluidine, irrespective of the analytical techniques used and hence was not characteristic of the other Gram-negative bacteria investigated. The developed methodology has the potential to be applied for clinical and food applications.
